A recent study was the first to document the use of tamoxifen for breast cancer prevention in women. The study was conducted at the Breast Cancer Research Foundation (BCRF) in Washington DC and the National Cancer Institute (NCI) in Bethesda. The research was published in theJournal of Clinical Oncologyin December 2016.
The study was conducted to evaluate the efficacy of tamoxifen in preventing the development of breast cancer in premenopausal women. The study was conducted in five different phases: Phase 1 (n=1,843), Phase 2 (n=1,843), 3rd (n=1,843), and 4th (n=1,843) stages. The women in each phase were randomly assigned to take either nolvadex (10mg) or tamoxifen (1mg) for five years. A total of 16 women (12-25 years of age) were enrolled per phase, with a total of 6 cycles of tamoxifen. The average number of cycles was 3.5. After a median follow-up of 7.5 years, there was an increase in breast cancer risk (odds ratio [OR], 1.20; 95% confidence interval [CI], 1.06-1.50; P=0.02) and a reduction in breast cancer mortality (OR, 1.31; 95% CI, 1.20-1.45; P=0.01) with nolvadex versus tamoxifen. The most common breast cancer risk was increased with nolvadex (OR, 1.34; 95% CI, 1.11-1.79; P=0.01). There was no statistically significant difference between nolvadex and tamoxifen in overall breast cancer risk (OR, 0.95; 95% CI, 0.78-1.21; P=0.42) and the overall risk of breast cancer (OR, 1.06; 95% CI, 1.02-1.22; P=0.85) in postmenopausal women. There was no statistically significant difference between nolvadex and tamoxifen in risk of breast cancer (OR, 0.99; 95% CI, 0.85-1.22; P=0.58) or overall breast cancer (OR, 0.89; 95% CI, 0.79-1.09; P=0.73).
Conclusion:Tamoxifen is not associated with a significant increased risk of breast cancer. Although it is not a clinically relevant drug, it should not be taken if a woman is breast cancer-free for longer than one year.
The results of this study provide new hope for breast cancer prevention. Although the risk of breast cancer is lower than other cancers, it is associated with a lower risk of breast cancer. This is important since the breast cancer risk was not changed in women who had tamoxifen, nor was the risk of breast cancer changed by any of the tamoxifen treatment groups. There is also a lack of research on the use of tamoxifen in the prevention of breast cancer. Further research is required to determine the efficacy and safety of tamoxifen in women with postmenopausal breast cancer.
ContraindicationsTamoxifen is contraindicated in patients with liver disease, a history of breast cancer, or a history of breast cancer. The safety and efficacy of tamoxifen in these patients has not been established.
Women with a history of breast cancer should not take tamoxifen, as the drug is likely to cause a decrease in estrogen levels.
